T cell exhaustion is the dysfunction of T cells caused by prolonged antigen stimulation. In chronic infections and cancer, T cells are exposed to persistent antigen and/or inflammatory signals.

There is a growing interest in reversing or overcoming the state of exhaustion, particularly in the production of CAR-T cells by including checkpoint inhibitor antibodies. The ability to simulate T cell exhaustion problem-solution scenarios is essential to provide further insight into this phenomenon.

This application highlights a multiplexed approach to assessing T cell exhaustion, cell health, and cytokine release in a single well.
